ANA Immunoblot (also called ANA Profile or ENA Profile by immunoblot) is a line immunoassay (LIA) that detects IgG antibodies against a panel of specific nuclear and cytoplasmic autoantigens associated with systemic autoimmune rheumatic diseases (SARDs). The panel typically includes antibodies against: dsDNA (double-stranded DNA), Sm (Smith antigen, core component of snRNP), SS-A/Ro 52 kD and 60 kD, SS-B/La, Scl-70 (topoisomerase I), CENP-B (centromere protein B), Jo-1 (histidyl-tRNA synthetase), PM-Scl, Mi-2, PCNA, ribosomal P protein, and histones. Each specificity is associated with particular conditions: anti-dsDNA and anti-Sm are highly specific for SLE; anti-SS-A/Ro and anti-SS-B/La for Sjögren's syndrome and neonatal lupus; anti-Scl-70 for diffuse cutaneous systemic sclerosis; anti-CENP-B for limited cutaneous SSc (CREST syndrome); anti-Jo-1 for polymyositis/dermatomyositis and antisynthetase syndrome.
